Probably some issues with your drive.

1) Generally helps to tell us what OS.
2) If it's XP/2K, open up windows explorer, right click on your C: drive, go into Properties, Tools tab, Error Checking, hit the check now button.
3) Reboot
4) Once everything comes back up, defrag.

If it's Me, 98, 95, run scan disk before you try to defrag.

It sounds like 2K or XP; I don't believe that Win9x has the option to schedule a CHKDSK, even though an older DOS version of the program is present on Win9x installs.

I'd open up a Command Prompt window, and at the prompt, type

CHKDSK C: /R

This is the full disk check that also checks for bad sectors on the hard disk. CHKDSK will tell you that it cannot lock the drive for access, and ask you if you'd like to check the drive on reboot. Hit Y and enter. Then reboot the system, wait 10-30 minutes while it checks, and try defragging on next reboot.
